- Abstract
Introduction: The present investigation highlights the formulation of buccoadhesive tablets of Ketotifen fumarate, a second-generation H1-antihistamine and mast cell stabilizer. The properties of Ketotifen may contribute to its antiallergic activity. Method: The mucoadhesive buccal tablets of Ketotifen fumarate were prepared by direct compression method using natural polymers like sodium alginate, xanthan gum, karaya gum, and gaur gum along with carbopol 934P in different ratios. Ethyl cellulose is used as a backing layer to promote the unidirectional flow of drugs in the buccal region. Results: After examining the flow properties of the powder blends the results were found to be within prescribed limits and indicated good flowing properties, hence it was subjected to compression. The tablets were evaluated for postcompression parameters like their appearance, thickness, hardness, friability, weight variation, swelling index, surface pH, drug content, mucoadhesive strength, and in-vitro release. Among the twenty trial formulations, the formulation FSA1 emerged as the promising formulation, based on the results of swelling index 71.89±2.48%, mucoadhesive strength 6.41±0.14 g and in-vitro drug release 69.15±1.10% within 8 hours. All the evaluation parameters gave a positive result and comply with the standards. Conclusion: The results indicated that the mucoadhesive buccal tablets of Ketotifen fumarate may be a good choice to bypass the extensive hepatic first-pass metabolism with an improvement in the bioavailability of Ketotifen fumarate through the buccal mucosa.
