PROTECTIVE EFFICACY OF NOVEL NANOPARTICLE BASED VACCINE AGAINST NEWCASTLE DISEASE.

- Abstract
Globally, Newcastle disease (ND) of poultry causes overwhelming economic losses. Although, live ND vaccines are administered to obtain effective prophylactic measures but all the requirements for improving infectivity and productivity in poultry industry may not be met. The present study attempts to improve both the health and immunity of animals using nanotechnology approaches. The aim of the study was to prepare and characterize copper oxide nanoparticles (CuONPs) using green chemistry principles. The prepared nanoparticle was used as an adjuvant with live ND vaccine to evaluate its immunogenicity in pre-challenged and post-challenged white leghorn chicks. CuO nanobars with an average particle size of 61 nm were prepared from aqueous extract of Momordica charantia fruits and conjugated with live ND vaccine. The adjuvancity levels of immunogenicity and protection against the R2B NDV strain were evaluated in 2-weeks old chicks after intramuscular immunization. The experimental groups were divided into 3 consisted of 15 animals each. The immune response (haemagglutinin inhibition, HI antibody titer), hematology and biochemistry were randomly assessed at different intervals. The results showed that the CuONP based nanovaccine has a higher survival rate with a protection rate of 80% with a slightly higher HI titer than the control and standard live vaccine. Furthermore, a significant change in hematological and biochemical parameters was observed. This is for the first time that CuONPs have been used in vaccine delivery. This work is expected to lay a foundation for future work on CuO nanomaterial to be used as drug delivery systems for vaccines.
