- Abstract
In order to study the genetic diversity of wild and farmed small yellow croaker populations, based on mtDNA Cytb gene and D-loop control area, two wild small yellow croaker populations and one cultured population (YZ) were collected from Zhoushan Shengsi sea area (SS) and Xiangshan Sanmenkou sea area (SMK). The genetic structure and genetic differentiation of Cytb were compared and analyzed. The sequence analysis showed that the Cytb gene sequence was 841 bp, its A+T content (50.2%) was similar to C+G content (49.8%); the D-loop region sequence was 629 bp ~635 bp, A+T content (58.9%) is much higher than C+G content (41.1%). The haplotype numbers of Cytb gene in SS, SMK and YZ populations are 26, 27 and 12, respectively, shared by SS and SMK populations 2 haplotypes (Hap1 and Hap13), and one haplotype (Hap41) shared by the SMK and YZ populations; the haplotype numbers in the D-loop region of the SS, SMK and YZ populations were 27, 30 and 10, respectively. The SMK population shared one haplotype (Hap4). The diversity analysis showed that the three populations belonged to high haplotype diversity (Hd>0.5). Among them, the SS and SMK populations had haplotype diversity and nucleotide The diversity is higher than that of the YZ population, indicating that the diversity of the wild population is slightly higher than that of the cultured population. The genetic differentiation index shows that the degree of differentiation between the two wild populations of small yellow croaker is very small, while there is moderate differentiation between the cultured population and the wild population. The genetic differentiation index The results of AMOVA and AMOVA analysis showed that individual variation within the population was the main source of genetic variation. The Tajima's D value and Fu and Li's value of the SS and SMK populations were negative in the Cytb gene and D-loop region sequence neutrality test results, and Cytb The Tajima's D value and Fu and Li's value of the gene were significantly (P<0.05) deviated from neutral, indicating that the two wild populations may have experienced population expansion. The haplotype phylogenetic tree showed that none of the SS, SMK and YZ populations exhibited The obvious geographical aggregation and the crossover among the groups indicated that the differentiation among the three groups was not obvious.
