- Abstract
Introduction: The role of adenosine and its receptors in acute pancreatitis remains unelucidated. Adenosine is endogenic nucleoside that participates in many important physiological processes and pathologies. It acts via specific membraneous receptors: P1, P2. The receptors are divided into 4 subtypes: A1, A2a, A2b, A3. The receptors A1 and A2 were detected in the pancreatic β islet cells. Aim of study: The purpose of the experiment was to examine the role of adenosine receptors in the course of sodium taurocholate-induced experimental acute pancreatitis (EAP) with special regard to receptor A1 and A2a antagonists. Material and methods: The experiments were performed on 50 male Wistar rats, subdivided into 5 groups: I - control rats (sham operated), II - the EAP group, III.1 - EAP group treated with the A1 antagonist DPCPX, III.2 - EAP group treated with the A1 selective antagonist 1,3-dipropyl-8-phenylxanthine, III.3 - EAP group treated with the A2a adenosine antagonist ZM 241385. The blood for α-amylase and lipase and tissues samples for the histological examinations for A1 and A2a receptor collected in 2,6,24 hours of the experiment. The intensity of inflammation was determined by biochemical scores (blood serum α-amylase and lipase activity) and histopathologically under light microscope and electron microscope and evaluating immunohistochemical response. Results and conclusions: The use of A2a receptor antagonist ZM 241385 enhanced the activity of the inflammatory process in comparison to group I but slightly decreased the activity in comparison to group II. Receptor A1 antagonists reduced histological scores of EAP in comparison to group II. The results of our experiment suggest that A1 and A2a adenosine receptors play a role in the course of sodium taurocholate-induced EAP. The antagonists of A1 and A2a receptors seem to alleviate the course of EAP.
