Optimization of microwave assisted extraction of flavonoids from raspberry with deep eutectic solvents and its activity.

- Abstract
Having raspberry dry powder as raw material, the flavonoid extraction yield as examining index, the microwave assisted extraction process of raspberry flavonoid with deep eutectic solvent was optimized, the optimal combination of deep eutectic solvents was screened, the effects of low eutectic solvent mole ratio, water content, liquid to solid ratio, microwave power, radiation time and reflux extraction time on the extraction effect were investigated. The extraction process was optimized by star-point response surface. The antioxidant activity of purified flavonoids was evaluated by determining the scavenging rate of DPPH and hydroxyl radical, and the effects on the growth of Escherichia coli, Staphylococcus aureus and Bacillus subtilis were investigated. The results showed that the extraction efficiency of choline chloride/sorbitol was better. The single factor test results showed that water content of deep eutectic solvents, liquid-solid ratio and microwave power were the main factors affecting the extraction of flavonoids. The optimal extraction process obtained by star-point response surface was as follows: Under the conditions of 1:2 choline chloride/sorbitol molar ratio, 31% water content, 38 mL/g liquid to solid ratio, 450 W microwave power, 150 s radiation time and 60 min extraction time, the extraction rate of flavonoids was 5.88%, and the relative error was 1.85% compared with predicted value. Antioxidant study results showed that the purified flavonoids had good antioxidant activity, and showed a dose-effect relationship with drug concentration. The results of antibacterial activity showed that flavonoids had certain inhibitory effect on the three common bacteria. In this study, microwave-assisted deep eutectic solvent extraction of flavonoids from raspberry had a good effect, and the extracted and purified flavonoids maintained good antioxidant and antibacterial activities.
