Effect of aqueous extract from coprinus comatus mycelium on alloxan induced diabetic mice.

- Abstract
Objective : To investigate the hypoglycemic effect and mechanism of Coprinus comatus mycelium water extract (JTGJST) on alloxan induced diabetic mice. Methods : The diabetic mice model was established by one time intraperitoneal injection of alloxan (160 mg/kg). Fifty diabetic mice were randomly divided into model control group, metformin hydrochloride group, JTGJST high-dose, mid-dose, low-dos group (750 mg/kg, 500 mg/kg, 250 mg/kg). Twenty normal mice were randomly divided into blank control group and JTGJST blank high-dose group. The mice were gavaged for 4 weeks, and blood glucose was measured every week. Then the blood glucose, glucose tolerance, liver glycogen, SOD and MDA were measured, and the liver and pancreas of mice were pathologically detected. Results : Compared with the model control group, each dose of JTGJST could significantly reduce the fasting blood glucose (P < 0.01), JTGJST mid-dose and high-dose group could enhance glucose tolerance (P < 0.01, P < 0.05), TGJST low-dose and middle-dose group could increase liver glycogen content (P < 0.05, P < 0.01) and SOD activity (P < 0.01, P < 0.05), each dose of JTGJST could reduce the content of MDA in liver (P < 0.01) and improve the damage of liver and pancreas to varying degrees. Conclusions : JTGJST can effectively reduce the blood glucose level of alloxan induced diabetic mice, protect liver and pancreas. The mechanism may be related to its antioxidant effect.
