- Abstract
Stachys bulgarica Degen & Neic (Bulgarian woundwort) is a Bulgarian endemic plant included in The Red Data Book of Bulgaria with conservational status "endangered". The plants from Stachys genus have been used in ethnomedicine for various inflammatory diseases and infected wounds. There are no data on in vitro multiplication of S. bulgarica and scarce information is available about its biological activity. In the present work we aim to develop an effective protocol for in vitro multiplication and ex situ conservation of S. bulgarica and comparative determination of the radical scavenging activity and phenolic composition of extracts isolated from in situ, in vitro cultivated and ex vitro adapted plants. In vitro shoot culture was induced from ripe dried seeds and multiplicated on basal MS medium. Ex vitro adaptation was accomplished in experimental field with 96% survival. The highest phenol and flavonoid quantity as well as antioxidant and radical-scavenging activity were observed in in situ and ex vitro adapted plants. Different growth conditions affect the phenol quantity and antioxidant potential of S. bulgarica. A collection of in vitro cultivated and ex vitro adapted plants was established which is an alternative approach for the preservation of this endemic species in Bulgaria.
