- Abstract
The objective of this study was to develop hydrocolloidal delivery systems based on chitosan and polyvinyl alcohol (PVA) that have been loaded with insulin for nasal administration, in considering the beneficial effects of insulin in diseases associated with the central nervous system (CNS), following nose-to-brain insulin delivery. This research was based on chitosan biodegradable, biocompatible, nontoxic characteristics, and the most important, the mucoadhesive properties and the capacity of the chitosan to enhance the penetration through the nasal mucosa of active pharmaceutical ingredients. The association of the chitosan with the synthetic polymer PVA increase the mucoadhesiveness and the residence time of the formulation on the mucosa. The evaluation of the hydrocolloidal delivery systems was based on an experimental design with the following formulation parameters: X1 = chitosan concentration, X2 = chitosan/PVA ratio and X3 = insulin loading. The identified response variables included surface characteristics and rheological profile. The findings suggested that chitosan-based hydrocolloidal delivery methods are suitable for nasal administration. Therefore, the formulated chitosan compositions have potential for further research.
