- Abstract
【Objective】The species and diversity of endophytic bacteria of Bletilla striata were studied, in order to provide references for better protection and management of B. striata. 【Method】The 16 S r DNA-V4 variable region of B.striata was sequenced by Illumina Mi Seq high-throughput sequencing technology. And using Qiime and Mothur softwares,the number of sequences and operational taxonomic units(OTUs) for each sample was sorted and calculated, so as to analyze abundance and distribution of species. 【Result】The results showed that, the numbers of effective sequences and OTUs for each sample were 49550 and 48, respectively. And the rarefaction curves showed that sequencing depth was adequate,and The number of OTUs was close to saturation. The endophytic bacteria of B. striata belonged to 8 genera viz., Sphingomonas(32.26%), Pedobacter(16.13%), Halomonas(16.13%), Agrobacterium(12.90%), Kaistobacter(6.45%), Shewanella(6.45%), Pseudomonas(6.45%), Brevundimonas(3.23%). 【Conclusion】The dominant endophytic bacteria species in B. striata belongs to Sphingomonas, Pedobacter, Halomonas and Agrobacterium. Furthermore, Illumina Mi Seq high-throughput sequencing technology can reflect accurately species composition and proportion, so it has obvious advantages and feasibility in the study of plant endophytic bacteria.
