The article focuses on the use of imaging-based sensors for the determination of mechanical forces exerted by cells. Topics include the use of force-sensitive fluorescence resonance energy transfer (FRET) sensors for the determination of the cellular force, construction of cell tethers by conjugating a ligand to DNA to tune the tether strength and evaluation of quenching of a conjugated fluorophore by quencher at the substrate.
