- Abstract
Objective To study the expressions of endothelin-1 (ET-1), endothelial nitric oxide synthase (eNOS) and neuronal nitric oxide synthase (nNOS) in penile tissues of the rats with compound stress type ED, and evaluatethe therapeutic effects of Yimusake extract and its composition, and explore its reproductive biological significance. Methods Total of 160 male SD rats, who were confirmed to have normal sexual function through mating experiment, were randomly divided into normal control group (N,15 rats) and the model group (M,145 rats). The rats in group M received environmental estrogen-like feed combined with cold stress compound intervention and to be a chronic compound stress rat model. After 35 weeks of modeling, ED models were screened from compound stress models through mating experiments and then randomly divided into ED model control group (ED), Yimusake group (Yimusake), alcohol extract group (AE), water extract group (WE), and combination of extracts group (COE). After 3 weeks intervention with the corresponding extracts and drugs, the expressions of ET-1, eNOS and nNOS in penile tissue were detected by immunohistochemistry and western blotting. Results (1) Results of immunohistochemistry: Compared with that in group N, the expression of ET-1 in group ED was significantly increased (P<0. 05), whereas the expressions of eNOS and nNOS were significantly decreased (P<0.05). After intervention, compared with that in group ED, the expressions of ET-1 in COE, WE and Yimusake groups were significantly decreased (P<0.05), while its expression in COE and WE groups were significantly lower than that in groupYimusakeand AE (P< 0.05). The expressions of eNOS and nNOS in group COE, AE, WE and Yimusake were also significantly higher than those in group ED (P< 0.05), but there was no significant difference in eNOS among the intervention groups (P> 0.05). The expression of nNOS in group AE and WE were significantly lower than that in group Yimusake (P<0.05), and its expression in group COE was clearly higher than that in group Yimusake, but there was no statistical significance (P>0.05).(2) Western blot results: Compared with that in group N, the ET-1 expression in group ED was significantly increased (P<0.001), whereas the expressions of eNOS and nNOS were significantly reduced (P<0.001). After intervention, the expression of ET-1 in group COE and WE was significantly lower than that in group ED (P< 0.001), and the expression of ET-1 in group COE and WE were also significantly lower than that in group Yimusake (P<0.001);The expression of eNOS in groups COE, WE, AE and Yimusake were significantly higher than that in group ED (P<0.001), and the expression of nNOS in groups COE, AE and Yimusake were significantly higher than that in group ED (P<0.001);Compared with those in group Yimusake, the expression levels of eNOS and nNOS in groups COE and AE had no significant difference (P>0.05), but their expressions in group WE were significantly lower than those in Yimusake group (P<0.001). Conclusion (1) The occurrence of ED rat model may be attributed to the increase of ET-1 and the decrease of eNOS and nNOS in the penile tissue, indicating that there exist endothelial dysfunction in this pathogenesis. (2) Yimusake extract and its composition can exert a therapeutic effect by regulating the levels of ET-1, nNOS and eNOS, and the regulating effect of COE is better than that of Yimusake tablets.
