基于网络药理学预测四逆汤抗心力衰竭的作用机制.

- Abstract
OBJECTIVE: To predict the mechanism of Sini decoction in anti-heart failure with the utilization of network pharmacology. METHODS: The Chinese herbal medicine database was searched to obtain the chemical constructions of Sini decoction related active ingredients; the database of active ingredients of Chinese herbal medicine and therapeutic target database was searched to screen the action target points of Sini decoction, so as to establish dataset of target points;the Online Mendelian Inheritance in Man database was searched to screen the related targets of heart failure; the protein to protein interaction network was used for constructing the interactive targets between Sini decoction and heart failure; the Cytoscape software was adopted to construct the interactive network chart of "ingredients-target points-diseases". RESULTS: 99 active ingredients, 176 directly active protein target points, 41 directly active protein target points of cardiovascular related disease and 489 interactive proteins were screened out, 8 of which were the common protein target points of Sini decoction and heart failure;totally 25 pathways were involved in the action of anti-heart failure, further investigation and survey had indicated that all the predicted pathways with higher relevancy were testified, such as kinase pathways of phosphatidylinositol 3-kinase-serine/threonine and mitogen activated protein, signal pathways of thyroid hormones and epidermal growth factor receptor ErbB , human cancer suppressor gene p53 pathways, etc. CONCLUSIONS: By mean of network pharmacology, this study has preliminarily revealed the multi-dimension regulatory network of the action of Sini decoction in anti-heart failure, so that provide theoretical bases for its clinical application.
