- Abstract
Objective To explore the possible mechanism of Hezhong zhiqiu granules in the treatment of allergic rhinitis (AR) based on the Toll-like receptor(TLR)- nuclear transcription factor-κB(NF-κB) pathway. Methods 12 AR patients (treatment group) admitted to the Department of Otolaryngology of Affiliated Traditional Chinese Medicine Hospital of Southwest Medical University from November 1,2017 to November 30,2018 and 5 healthy volunteers (healthy control group) during the same period were selected as the research objects. Patients with AR were given oral and discontinued treatment with Hezhong zhiqiu granules, and healthy volunteers were not given any intervention. The venous blood and mucosal tissues of the nasal cavity and nasal mound region were collected from patients before and after treatment and healthy volunteers only before treatment, and stored in a refrigerator at -80 ℃. Enzyme-linked immunosorbent assay was used to detect the expression levels of interferon-γ (IFN-γ) and interleukin-4 (IL-4) in venous blood. Fluorescence quantitative polymerase chain reaction and western blotting were used to detect the RNA and protein changes of TLR4 and NF-κB in mucosal tissues in the nasal cavity and nasal collum area. Results Compared with the healthy control group, the expression level of IFN-γ in the serum of patients in the treatment group before treatment was down-regulated, and the expression level of IL-4 was up-regulated, and the differences were statistically significant (P <0.05). TLR4 in the nasal cavity and nasal colloid mucosa of the treatment group and NF-κB mRNA gene protein expression levels were up-regulated at the same time, the differences were statistically significant (P <0.05). After treatment, the above three indicators of patients in the treatment group were negatively correlated with those before treatment (P <0.05). Conclusion Hezhongzhiqiu granules may be able to treat AR by regulating the TLR4-NF-κB pathway.
