- Abstract
Objective: This study was carried out for the detection and molecular characterization of important virus diseases in canola fields of Canakkale and Tekirdağ provinces in Turkey. Material and Methods: Canola plants were examined visually and ones showing virus and virus-like symptoms were collected. Collected samples were tested with DAS-ELISA to determine the presence of turnip mosaic virus (TuMV), cucumber mosaic virus (CMV) and cauliflower mosaic virus (CaMV). Two isolates were selected for each virus and the desired gene regions were partially amplified, cloned and sequenced. This sequences obtained were used in similarity and phylogenetic analysis. Results: As a result of the tests, 16 and 3 samples were found to be infected with TuMV and CMV, respectively. In one example, CMV and TuMV mixed infection was detected. None of the samples collected had CaMV infection. In the similarity analyses, TuMV isolates showed similarities 80-94% and 90-98%, CMV isolates showed similarities 77- 98% and 82-100% with world isolates at nucleotide and amino acid levels, respectively. Moreover in phylogenetic analyses, TuMV and CMV isolates were found in the Asian BR and IA groups, respectively. Conclusion: TuMV and CMV infections in canola production areas were detected and partially amplified gene regions molecularly characterized with this study.
