- Abstract
Sperm antibodies play an important role in cases of unexplained infertility. Hence, the availability of a test for routine detection of those antibodies is highly desirable. Therefore an ELISA for the detection of sperm antibodies in human sera was developed. Basis of the ELISA was a soluble extract of spermatozoa/ surface antigens from pooled spermatozoa. With this antigen preparation, wells of microtiter plates were coated. After saturation of remaining protein binding capacity by horse serum, sperm antibodies against these antigens were specifically bound. Their detection was achieved by peroxidase coupled anti human-IgG-antiserum as second antibody. For validation of the sperm antibody-ELISA three groups of sera were utilised: sera from children, pregnant women and unexplained infertile patients. The sera of children as well as of pregnant women were regarded as negative control groups. This allowed definition of a threshold value, above which a human serum was classified sperm antibody positive. In a group of 137 unexplained infertile patients, 22 sperm antibody positives, corresponding to 16%, were detected. The sperm antibody-ELISA developed turned out to be practical and reproducible. The test is now routinely employed to diagnose immunological aspects of unexplained infertility.
