- Abstract
Aphids are among the most serious agricultural pests of tropical, subtropical and temperate regions. They infest almost every plant by feeding on its phloem (Clark et al, 1997) and acts as vectors of plant viruses. They constitute invasive threat to native and cultivated plants and cause very severe damage to crops. The reliable and accurate identification of aphid species is important for the effective integrated pest management strategies. However, the accurate identification of aphid is difficult because many species are morphologically similar and have higher level of intraspecific variation. The present paper deals with the identification and amplification of ten aphid species viz., Aphis gossypii, Aphis craccivora, Dactynotus compositae, Rhopalosiphum nymphaeae, Aphis nerii, Myzus persicae, Lipaphis erysimi, Toxoptera citricidus, Macrosiphum rosaeformis and Macrosiphonella sanborni. In the present study, polymerase chain reaction (PCR)-sequencing based on DNA barcoding and SSR were applied to identify aphid species collected from different areas of Delhi and its NCR. As both PCR and DNA barcoding provide quick and accurate tools for identification of these aphid species.
