Effects of Hyperglycemia Induced by Type 2 Diabetes on Bone Strength,Bone Mineral Density, and Bone Quality in Diabetic Mice.

- Abstract
The aim of this study was to elucidate the effects of persistent hyperglycemia induced by type 2 diabetes on bone metabolism by examining the bone strength, bone mineral density (BMD), and bone quality of BKS.Cg-+ Leprdb!+ Lepr/Jcl (db/db) mice,an animal model of obese type 2 diabetes. Five-week-old db/db mice and age-matched non-diabetic (BKS.Cg-zw +/+ Lepr/Jcl, db/+m) mice, as the control, maintained until 28 weeks of age, after which their blood and lower limb bones were collected. BMD was measured using quantitative computed tomography, and serum biochemical markers were examined. Bone strength was evaluated using a 3-point bending method, and bone histomorphometry was analyzed. Compared with the control mice, the db/db mice showed decreased serum osteocalcin, a bone formation marker, and significantly increased serum tartrate-resistant acid phosphatase-5b, a bone resorption marker. Also, the trabecular and cortical BMD of the femur and tibia were significantly reduced in the db/db mice. This deterioration of bone metabolism in the db/db mice was accompanied by a decrease in the femoral bone strength properties. Bone histomorphometric analysis revealed the impaired microstructure of trabecular bone in the proximal tibia metaphysis in db/db mice. Moreover, osteoblast surface and mineralizing surface were decreased, whereas osteoclast surface and number were increased in db/db mice. These results indicate that type 2 diabetes-induced bone fragility is caused by a decrease in the BMD and a deterioration of the bone quality.
