- Abstract
Background Cytochrome P450 superfamily genes are involved CYP1A2, CYP2C9, CYP2C19, CYP2D6 and CYP3A4, which is responsible for the metabolism and elimination of approximately 80% of clinically use drugs. Rapidly genotyping of the most prevalent alleles is goal. Methods The primers are designed and set up for thirty main variants of the five genes and were described using two quick and accurate methods (tetra ARMS PCR and ASA). These methods were used to statistics measurement of three Iranian ethnic groups in order to obtain a population estimate of these variants in the population of Iran. One hundred and fifty Iranian healthy volunteers (70 Fars, 50 Azari and 30 Kurd) were selected in this study. After DNA extraction, these genotyping methods were used for the variants of CYP1A2, CYP2C9, CYP2C19, CYP2D6 and CYP3A4 genes. Results In this study, Genotype frequency of wild, heterozygote and mutant of CYP1A2 *4, CYP2C9 *2 & *3, CYP2C19 *2, *3 & *17, CYP2D6 *2, *4, *10,*35 & *41, CYP1A2 *4 variants with P<0.05 was exposed a statistically significant difference. The rest of the variants had zero frequency. Conclusion These results lead to increase risk factors for using several drugs and the patients should be precautious of the side effect of these drugs (Caffeine, Aflatoxin B1, Acetaminophen, Warfarin, Phenytoin, Acenocoumarol, Tolbutamide, nonsteroidal Clopidogrel, Omeprazole, Mephenytoin, Proguanil, Diazepam Fluoxetine, Haloperidol, Resperidone, Codeine and Erythromycin). These techniques (tetra ARMS PCR and ASA) are very suitable for cases that need the quick and accurate answers. Only one day is enough for each case to determine the type of 30 variants.
