- Abstract
Lentinus edodes was used to extract lentinan (LNT) by double enzyme coupling ultra high pressure extraction method. The extraction process was optimized by response surface methodology on the basis of single factor experiment. The antioxidant activity was also studied. The results showed that the optimum extraction conditions were: the amount of double enzyme (cellulase: papain, mass ratio 1:2) 2.5%, pH of enzymatic hydrolysis 5.5 and reaction pressure 450 MPa. Under these conditions, the extraction rate of LNT was 8.83%. LNT had a certain scavenging capacity for DPPH radicals and hydroxyl radicals. When the concentration of LNT was 25mg/mL, the scavenging rates of DPPH radical and hydroxyl radical were 79.62% and 81.59% respectively. This study provides a new extraction method for LNT.
