Effect of resveratrol on inflammatory reaction after spinal cord injury in rats.

- Abstract
Objective To study the effect of resveratrol on inflammatory reaction after spinal cord injury in Rats. Methods A total of 63 rats were randomly assigned into three groups with 21 in each: sham group, control group and resveratrol group. Rats in sham group underwent operation exposure only; the model of spinal cord injury was established in control group by Allen's weight hit. Resveratrol group received intraperitoneal injection of resveratrol at the dose of 200 mg/kg. At 24, 48 and 72 h after operation, the contents of interleukin-1β (IL-1β), interleukin-10 (IL-10) and tumor necrosss factor-β (TNF-β) were observed by ELISA assay. The activity of myeloperoxidase (MPO) was observed by colorimetric method assay. At 72 h, the expressions of IL-1β, IL-10, TNF-β and MPO in spinal cord injury tissues were observed by immunohistochemical staining and Western blot assay. Results IL-1β, IL-10 and TNF-α expressions as well as MPO actmty were significantly lower at 24, 48 and 72 h in resveratrol group than in control group (P<0.05). The results of colorimetric method showed that resveratrol group had significantly downregulation MPO activity comparied with control group at 24, 48, and 72 hours (P< 0.05). The resutts of immunohistochemical staining and Western blot assay showed that IL-1β, IL-10, TNF-α and MPO expressions were signicantly decreased in resveratrol group compared with those in control group at 72 h after operation (P<0.05). Conclusion Resveratrol has a protective effect on spinal cord injury in rats by reducing inflammatory mediator activity after spinal cord injury.
