- Abstract
Methotrexate is a folic acid analog and a specific inhibitor of dihydrofolate reductase. It binds very avidly to the enzyme. The binding is due to its diaminopteridine structure. The amoni group affect hydrogen bonding with the active site of the enzyme whereas, the natural substrate folate or dihydrofolate has an amino group on position 2 and a hydroxyl group at position 4 on the pteridine ring. The hydoxyl group exists as a keto group. When dihydrofolate reductase is inhibited by methotrexate it cannot carry out its normal function which is to convert folate to dihydrofolate.
