The author reflects on the use of Escherichia coli as host organism for recombinant protein production. Topics discussed include simplification of cloning and gene expression processes due to availability of plasmid expression vectors, exploitation of site-specific and homologous recombination pathways for gene integration in the genome of the organism, role of clustered regularly interspaced short palindromic repeats (CRISPR) system in bacteria to detect and degrade foreign nucleic acids.
