IL-32 及 IL-5 对哮喘小鼠气道黏液高分泌的影响及地塞米松的干预作用.

- Abstract
Objective To investigate the influence of IL-32 and IL-5 on airway mucus hypersecretion in asthmatic mice and dexamethasone(DEX) interventional effect. Methods Thirty clean class male BALB/c mice were randomly divided into the normal saline(NS) control group(NS group),asthmatic model(AS) group(AS group) and AS+DEX intervention group(AS+DEX group), 10 cases in each group. The AS group was given ovalbumin(OVA) suspension fluid 0.2 mL by intraperitoneal injection on1,13 d for causing sensitization and 5% OVA solution on 19 d for conducting atomization stimulation, 30 min per time for successive 5 d. The AS+DEX group was interfered by intraperitoneal injection of DEX 2 mg/kg at 30 min before atomization stimulation for successive 5 d. The remaining was same to that in the AS group. The NS group was given normal saline for causing sensitization and stimulation. The levels of cytokine IL-32 absor bance(OD) and IL-5 content in bronchoalveolar lavage fluid(BALF) were determined by ELISA, the lung tissues were observed by hematoxylin-eosin(HE) staining and alcian blue/periodic acid Schiff(ABPAS) staining, the expression of Muc5ac mRNA in lung tissue was detected by RT-PCR, the expressions of mucin 5ac(Muc5ac)and IL-32 protein were measured by using the immunohistochemical method. Results The IL-5 content and IL-32 OD, AB-PAS positive expression area, and expressions of Muc5ac mRNA, Muc5ac and IL-32 protein in BALF of the AS group were all significantly increased compared with those in the NS group, while the above indicators in the AS +DEX group were significantly decreased compared with the AS group, but which were still higher than those in the NS group, the differences were statistically significant(P<0.05). Conclusion IL-32 and IL-5 can promote airway Muc5ac protein high expression in mice with asthma, and dexamethasone may inhibit mucous hypersecretion by down-regulating their expressions.
