- Abstract
Objective To investigate the effect of curcumin on nuclear factor erythroid 2-related factor 2 (Nrf2) expression in testicular tissues of prepubertal rats with unilateral testicular torsion. Methods Total of 32 four-week-old healthy male SD rats were randomly divided into four groups including sham operation group, saline group, curcumin single-dose group, and curcumin successive group, and 8 rats in each group. The animal model of left testicular torsion reduction (720° for 2h) was first established, and then were treated with medication. After six weeks of surgery, all the rats were executed and their left testes were collected immediately for detecting the activity of superoxide dismutase (SOD) and the content of malondialdehyde (MDA), and the protein expression of Nrf2 and its nuclear translocation rate. Results The activity of SOD was decreased and the content of MDA was increased when testicular tissue suffered from ischemia-reperfusion injury. The activity of SOD was increased and the content of MDA was decreased in curcumin single-does and successive groups (P <0. 05). The nuclear translocation rate of Nrf2 was increased when testicular tissue suffered from ischemia-reperfusion injury. The nuclear translocation rate of Nrf2 in the curcumin successive group increased more significantly than that in the curcumin single-dose group (P<0.05). Conclusion Curcumin has an obvious long-term protective effect on testis with ischemia-reperfusion injury through activating the signal pathway of Nrf2/ ARE, protecting the activity of SOD and inhibiting lipid peroxidation. The protective effect of the curcumin successive group is better than that of the curcumin single-does group.
