- Abstract
The experiment was to screen β-glucosidase high yielding strains from nature, identify them and optimize the enzyme production conditions. The β-glucosidase strains were isolated and screened by aesculin chromogenic technique, and enzyme production conditions were optimized by solid fermentation technique. The β -glucosidase strains were identified by morphological characteristics and phylogenetic analysis. The results showed that a β-glucosidase producing strain was screened from the karst natural vegetation area of Hechi, Guangxi, and identified as Aspergillus aculeatus. The optimal enzyme producing conditions were pH 7.0, temperature 28 ℃ and liquid loading of 40 mL/250 mL. After optimization, the enzyme production capacity of the Aspergillus aculeatus strain reached 42.75 U/mL, which was 10.9 times higher than the starting condition. The study indicates that Aspergillus aculeatus screened in the study can produce β-glucosidase stably, and the enzyme-producing capacity is greatly improved after optimization by solid fermentation, with the highest enzyme production under neutral conditions, indicating its application value in feed production.
