16S rRNA ribotyping Streptococcus mutans from dental caries patients.

- Abstract
Background: Streptococcus mutans is one of the most common microbes responsible for the development of an infective, multifactorial disease affecting the hard tissue dental caries. It results due to an imbalance in the biofilm community creating an acidic environment. To analyze the genetic polymorphism of S. mutans, 16S rRNA-based ribotyping method is adopted and is completely reliable for the bacterial community analysis. The objective of this study is to evaluate and observe the different ribotyping patterns seen in S. mutans from dental caries patients. Materials and Methods: Saliva samples were collected from 20 children from 6 to 10 years of age. Following processing and bacterial culture of S. mutans, the isolates were obtained. This was followed by amplification of the samples in a thermocycler under suitable polymerase chain reaction (PCR) conditions. Amplification and sequencing of the 16S rRNA were performed to confirm bacterial identification, and then, the data analysis was performed using the Statistical Software SPSS. Results: Streptococcus sequences in Gene Bank revealed the putative amplification of a 517 bp DNA fragment of the glucosyltransferase gene exclusively in five S. mutans strains. 12 of 20 strains were identified by 16S rRNA sequence analysis with 100% similarity by targeting Eubac 27F primer. Conclusion: 16S rRNA and gene sequences are remarkable tools in the identification of different types of species, and PCR ribotyping analysis is reliable for the characterization of S. mutans.
