Effect of Methamphetamine on the Small Intestinal Morphology of Rats.

- Abstract
Objective: Objective of the study was to investigate the effects of long term Methamphetamine administration on the small intestinal morphology in male Wister rats. Materials and Methods: Thirty six (36) rats were divided into control (n = 9), placebo (n = 9) and Methamphetamine (MA, n = 18) groups. Placebo and MA groups received intraperitoneal injection of either saline or 5 mg MA for 12 weeks. After 12 weeks, one third of the rats from control and placebo groups and 8 rats from MA group were sacrificed. Remaining rats of MA group were then divided into 2 subgroups, withdrawal week 2 and withdrawal week 8 and were sacrificed 2 weeks and 8 weeks along with control and placebo after the stoppage of MA injection. Body weight was recorded. Ileum was collected from each rat, fixed in formalin, paraffin embedded tissue sections were cut in microtome and Periodic acid Schiff stain was done to observe intestinal morphology. Results: Body weight of the MA group decreased significantly as compared to the control group (336 ± 21 vs 390 ± 26, p < .001). 8 weeks after withdrawal of MA, body weight increased significantly (390 ± 26 vs 419 ± 26, p < .000). Villus height decreased in MA group as compared to control and placebo but was not significant. 2 weeks after withdrawal of MA, villus height increased significantly as compared to MA group (623 ± 171 vs 242 ± 20 micron, p < .000). Crypt depth and the number of goblet cells in villus did not differ significantly among the groups. Goblet cells of the, crypt increased significantly in MA group as compared to placebo (20 ± 2.7 vs 14.7 ± 1.8, p < .001). 2 weeks after withdrawal of MA, goblet cells decreased in number as compared to MA group (14.4 ± 3.1 vs 20 ± 2.7, p < .004). Conclusion: Increased number of goblet cells were observed to compensate the compromised villus height to maintain the intestinal barrier in MA group.
