A Molecular Phylogenetic Study of 10 Polytrichaceae Plants Using ISSR Markers.

- Abstract
In this study, genomic DNA of 10 Polytrichaceae plants were successfully extracted with modified CTAB method to establish an appropriate PCR reaction system of Polytrichaceae. ISSR molecular markers were used to analyze the genetic diversity among 10 Polytrichaceae plants. According to the results, a total of 7 ISSR primers were identified with polymorphism. By using the screened ISSR primers, 59 bands were amplified, including 36 polymorphic bands, and the percentage of polymorphic bands (PPB) was 61.02%. Based on the results of ISSR analysis, experimental materials were divided into two categories by UPGMA cluster analysis; Polytrichastrum longisetum (Sw. ex Brid.) G. Sm. was clustered into one category, and other nine Polytrichaceae plants were clustered into one category, which was basically similar to their taxonomic status. This study indicated that ISSR molecular markers could be used to investigate the phylogenetics of Polytrichaceae plants.
