Apoptotic effect of quercetin on human colon cancer cell line SW-480.

- Abstract
Cancer is the second cause of death and colon cancer is the most prevalent cancer worldwide so treatment alternatives are always on the agenda. The etiology of human colon cancer is very complex, but diet is considered to be one of the important reason of it and researches think that lots of people are at risk of colon cancer because diet style. In light of this information, we aimed to investigate whether Quercetin (Que) has apoptotic effect on SW-480 colon cancer cells. Quercetin is a type of bioflavonoid naturally occurring in various edible plants. Also it has antiproliferative effects. The SW-480 colon cancer cell line was cultured in monolayer model. Cells were treated with Que at 24, 48 and 72 hours of incubation. TUNEL assay were used to determine the apoptotic cells in the monolayer culture. The BrdU labeling index was used An IC 50 inhibition dose of Que in SW-480 colon cancer cell was 200 μM/ml at 24, 48 and 72 hours of incubation. The control group had a normal pattern of Sphase fraction and many of the SW-480 cells nuclei were observed to be positive for BrdU. TUNEL positive cells were detected after treatment with Que in the monolayer cultures. The dead cell count was higher in the SW-480 cell lines with Que applied than in the control. We concluded that Que inhibit colon cancer growth by apoptosis with use caspase pathway. Further in vivo studies are needed to confirm our findings in humans. Three-dimensional studies, caspase analyses and AIF immunestaining analyses are still ongoing.
