- Abstract
To increase total flavonoids in Phellinusbaumii mycelia, protoplasts from the wild strain were treated by the room temperature plasma(ARTP)mutagenesis. Three resultant strains, A67, A130, and A157, showed both high flavonoids yields and genetic stability after five consecutive subcultures. Compared to the control strain SH1, total flavonoids content of A67, A130, A157 strains increased by 86.7%, 20.0% and 60.0%, respectively. The antagonistic experiment and the RAPD analysis indicated genetic differences between the three selected mutants and the control strain. Compared to the control strain, the ethanol extracts of A67 mycelia exhibited stronger antioxidant activities in vitro, contained higher Trolox equivalent antioxidant capacity(TEAC)and ferric reducing ability of plasma(FRAP)values. The results of this study suggested that ARTP-induction is an efficient and practical method for Phellinus strain improvement and thus provided a basis for the development and application of P. baumii flavonoids related products.
