Growth-Inhibitory Effect on Cancer Cells of the Extract from Crotalaria Sessiliflora L. and Identification of Anticancer Agents.

- Abstract
In this study, with the extracts from different parts of Crotalaria sessiliflora L., we tested their cytotoxicity in and growth-inhibitory effect on cancer cells through CytoScanTM-TPM cytotoxicity and MTT assays and structurally identified unknown materials. The CytoScanTM-TPM cytotoxicii:y assay found that the extracts from leaves, roots, and stems show concentration-dependent significant differences (p<0.05, p<0.01) in the cell-survival rate compared with the control group; and the MTT assay indicated that for the extract from leaves there is a statistically significant decrease (p<0.01) in the cell-survival rate. Based on these findings, we collected and quantified a unknown peak, but only known as an effective component of Crotalaria sessiliflora L., using a Prep-HPLC system and obtained a molecular weight of 325.35 from the quantified peak as determined by GC/MS. By means of 1H-NMR and 13C-NMR measurements, the peak was identified as monocrotaline with a chemical formula of C16H23NO6, where 23 protons and 16 carbons are included. When quantified by HPLC, the monocrotaline content was 9.18mg/100g in leaves, 9.30mg/100g in steams, 6.70mg/100g in roots, and 8.86mg/100g in seeds; As stated, the highest level was measured in stems. All these results suggest that further studies of Crotalaria sessiliflora L. will reveal its availability as both a natural antioxidant and an anticancer agent and its potential for prevention and treatment of cancer.
