- Abstract
The carboxyl, active hydrogen and hydroxyl in citrinin (CIT) structure were conjugated with bovine serum albumin (BSA), ovalbumin (OVA) and keyhole limpet hemocyanin (KLH) to prepare complete antigens. After mice were immunized, the paired antiserum and coating antigen with optimal competitive reaction were screened using indirect competitive enzyme-linked immunosorbent assay (IC-ELISA), and the IC-ELISA for rapid detection of citrinin in wine was constructed. The results showed that the optimal antiserum and coating antigen was the serum of immunization CT-ICLH (H) and CT-BSA (H), respectively. The detection linear range of the IC-ELISA method was 0.1-100 μg/L (R²= 0.969 88), and the detection limit was 0.1 µg/L. The half inhibitory concentration (IC50) value of citrinin antiserum was 9.8 µg/L, and the antiserum had no cross reactivity to salbutamol, ractopamine, patulin and aflatoxin Bl. The standard recovery rate for the detection of CIT in wine by the method was 92.01%-119.33%, the relative standard deviation (RSD) of standard recovery rate test results was 2.35%-5.22%. 14% ethanol had no significant effect on the detection results of IC-ELISA (P>0.05), indicating the residual citrinin in wine without pretreatment could be quickly and directly detected.
