- Abstract
Objective: To investigate the effects of WSTF on the proliferative and invasive abilities of lung cancer cells. Methods: WSTF stably overexpressed A549 cell line (A549-WSTF) and control A549 cell line (A549-control) were established by lentiviral infection. Proliferation assay and colony formation assay were used to observe the effects of WSTF overexpression on the proliferative ability of lung cancer cells. Trans-well migration assay and invasion assay were performed to study the influence of WSTF on invasive capacity of lung cancer cells. Results: WSTF protein level in A549-WSTF cells was significantly higher than that of control cells (P=0. 0004). WSTF overexpression significantly promotes proliferation (P=0.002, 0.0004, 0.0002 and 3.21× 10-5 on day 1 to day 4) and colony formation abilities (P=0.004) of lung cancer A549 cells. In addition, WSTF overexpression increases the migration ability of lung cancer cells indicated by migrated cell OD570 value, with A549-WSTF (0.626± 0.013) much higher than A549-control (0.322± 0.010) (P=2. 37× 10-5). Similar results were observed in trans-well invasion assay with the OD570 value of A549-WSTF (0.600± 0.027) much higher than that of A549-control (0.333± 0.017) (P=0.0004). Conclusion: WSTF may act as an oncoprotein by promoting the proliferative and invasive abilities of lung cancer A549 cells, and thus could be a promising therapeutic target for the clinical treatment of lung cancer.
