Role of Transforming Growth Factor-Beta Signaling Modulators in Periodontal Disease.

- Abstract
Transforming growth factor beta 1 (TGF-β1) plays a crucial role in protection of many organs from inflammation. TGF-β1 is expressed as a latent cytokine and needs to be activated for function. Epithelial integrin avβ6 and thrombospondin-1 (TSP-1) serve as activators of TGF-β1. Mice deficient of β6 integrin (β6-/-) develop periodontal disease with apical migration of junctional epithelium, inflammation and bone loss. The aim of the present investigation was to find out whether TSP-1 jointly with avβ6 integrin participates in TGF-β1 activation and protection of periodontium from inflammation. In addition, we hypothesized that the TGF-β1-mediated immunoprotection is mediated via Smad3, a key signalling molecule in the TGF- beta;1 pathway.
