- Abstract
One of the important processes in histological preparations is fixation. Fixation aims to keep cells, tissues, and their components unchanged so that the structure can be maintained as when the cell is alive and prevent autolysis. This literature review aims to determine the use of fixative solutions that are suitable for several macromolecules such as proteins, lipids, carbohydrates, and nucleic acids. Literature review data in the form of secondary data obtained from books, journals, or articles related to fixative solutions without limitation of publication year. The database was sourced from Google Scholar, PubMed, NCBI, Science Direct, Springerlink, Research Gate, and Nature. The keywords for searching books are tissue processing, pathology, and histology. Keywords for journal or article searches are protein fixation, formaldehyde fixation lipid, fixative for nucleic acid, glyo-fixx fixative, mercury chloride fixative lipid, formalin fixed carbohydrate, and ethanol or methanol for fixative lipid. Based on the literature review, it is known that the recommended fixatives for protein are glyo-fixx and HOPE, fixatives for lipid are osmium tetroxide and Champy's fluid, fixatives for carbohydrate are 80% ethanol, Bouin solution and Gendre's fluid, fixatives for nucleic acid are EDTA solution, carnoy, methacarn and Bouin fixatives. It can be concluded that each macromolecule has different properties, thus the selection of fixative solution should meet the properties of the macromolecule to be observed.
