Antioxidant effect of grape seed and Vitamin C on D-galactose model mice.

- Abstract
Objective : To study effects of grape seed extract (GSE), vitamin C (Vc) and grape seed vitamin C capsule on antioxidant ability in D-galactose model mice. Methods : Except blank control group, other mice were intraperitoneal injected with d-galactose for 6 weeks. Based on MDA level, mice were randomly divided into model group, low-dose vitamin C group, high-dose vitamin C group (8.3, 16.7 mg/ kg · bw), grape seed extract low-dose and high-dose groups (8.3, 16.7 mg/ kg · bw), grape seed vitamin C capsule low-dose and high-dose groups (50, 100 mg/ kg · bw). The content of malondialdehyde (MDA), reduced glutathione (GSH), glutathione peroxidase (GSH-PX) and antioxidant dismutase (SOD) levels in brain, liver and blood were determined after 30 days of continuous administration. Results : Compared with model group, liver coefficient was decreased and spleen coefficient was increased in Vc high-dose group, GSE high-dose group, grape seed Vc capsule low-dose group and high-dose group (P < 0.01, P < 0.05). The thymus coefficient of grape seed Vc capsule high-dose group was significantly increased (P < 0.01). The values of GSH, GSH-Px and SOD in high-dose vitamin C group, high- dose grape seed extract (GSE) group and high and low-dose grape seed vitamin C capsule group were increased, while the value of MDA was decreased. The differences were significant compared with the model group (P < 0.01, P < 0.05). Conclusion: Vitamin C, grape seed extract and grape seed vitamin C capsule can restore the oxidative damage of d-galactose aging mice and improve antioxidant enzyme activity.
