- Abstract
Background: Staphylococcus aureus is a microorganism of great importance in food, it is a major cause of contamination in dairy industries and the its accompanying microbiota can create microenvironments that favor the ability of S. aureus to survive under adverse conditions. It has recently been shown that the capacity of adhesion and invasion of epithelial cells of such microorganisms is a key process for the pathogen survival and colonization of epithelial tissues and to cause disease. Objectives: To evaluate in vitro S. aureus and its accompanying microbiota isolated from Minas cheese for the ability of adhesion and invasion of Hep-2 cells. Methods: S. aureus and accompanying microbiota (Enterococcus faecium) were placed at a final concentration of 1x 107 CFU/mL in wells containing the Hep-2 cells with a concentration of 5 x 105 cells per well. Individual experiments of adhesion and invasion were carried out to determine the total associated bacteria (adherent and invasive) for each of the microorganisms. For invasion experiments, the same methodology was used, with the addition of gentamicin (2mg/mL) to remove adherent bacteria and thus, to quantify CFUs of invasive bacterial cells. The strains were also evaluated for adherent and invasive capabilities in co-culture. Control experiments were done for growth of each strain in each media used. Results: S. aureus and E. faecium showed a remarkable capacity for adhesion and invasion of Hep-2 cells when tested individually. S. aureus showed a capacity of adhesion of 105 CFU and invasion of 104 CFU and, for E. faecium the capacity for adhesion and invasion were 104 and 103 CFU, respectively. No significant difference was observed in the adhesion and invasion of microorganisms evaluated in co-culture, compared to single cultures. Conclusions: S. aureus and E. faecium showed adhesion and invasion capacity in Hep-2 cells and, co-culture of the isolates did not alter the virulence potential of the microorganisms evaluated.
