Paenibacillus polymyxa produces fusaricidin-type antifungal antibiotics active against Leptosphaeria maculans, the causative agent of blackleg disease of canola.

- Abstract
A bacterial isolate capable of inhibiting the growth of Leptosphaeria maculans (Desmaz.) Ces. & De Not., the causative agent of blackleg disease of canola (Brassica napus L. and Brassica rapa L.), was identified as a potential biological control agent. This environmental isolate was determined to be Paenibacillus polymyxa based on its (i) biochemical and growth characteristics and (ii) 16S rRNA sequence similarity, and was given the strain designation PKB1. Antifungal peptides were produced by P. polymyxa PKB1 around the onset of sporulation, with optimal production on potato dextrose broth. The antifungal peptides were extracted from P. polymyxa PKB1 cells and (or) spores using methanol and were purified using size exclusion and reverse-phase chromatography. Characterization of the antifungal peptides was done using amino acid compositional analysis, Edman degradation sequencing from partially hydrolyzed material, and a variety of mass spectrometric methods. The purified antifungal material was found to be a mixture of related peptides of molecular masses 883, 897, 948, and 961 Da, with the most likely structure of the 897-Da component determined to be a cyclic depsipeptide with an unusual 15-guanidino-3-hydroxypentadecanoic acid moiety bound to a free amino group. These compounds are therefore members of the fusaricidin group of cyclic depsipeptides.Key words: cyclic depsipeptide, biocontrol agent, allo-threonine.Un isolat bactérien capable d'inhiber la croissance de Leptosphaeria maculans (Desmaz.) Ces. & De Not., l'agent causal de la jambe noire du canola (Brassica napus L. and Brassica rapa L.), a été jugé comme un agent potentiellement utile dans la lutte biologique. Cet isolat environnemental a été identifié Paenibacillus polymyxa d'après (i) ses caractéristiques biochimiques et de croissance et (ii) la similitude de la séquence de l'ARNr 16S et il a été désigné souche PKB1. Les peptides antifongiques de P. polymyxa PKB1 sont produits au début de la phase de sporulation et leur production est optimale dans un bouillon pomme de terre-dextrose. Ces peptides antifongiques ont été extraits de cellules et (ou) spores de P. polymyxa PKB1 par un procédé au méthanol et purifiés par tamisage moléculaire et chromatographie de phase inversée. La caractérisation de ces peptides antifongiques a été obtenue par une analyse de la composition en acides aminés, séquençage après dégradation d'Edman sur du matériel partiellement hydrolysé et une variété de méthodes de spectrométrie de masse. Le matériel antifongique purifié s'est révélé comme étant un mélange de peptides apparentés de poids moléculaires de 883, 897, 948 et 961 Da et il semble que la structure la plus vraisemblable du composé de 897 Da soit un depsipeptide cyclique possédant une moitié peu fréquente d'acide 15-guanidino-3-hydroxypentadécanoïque liée à un groupement aminé libre.
