- Abstract
Effect of rhizobacteria containing ACC-deaminase (1-aminocyclopropane-1-carboxylate deaminase) activity was studied in the Department of Soil and Environmental Sciences, University of Agriculture, Faisalabad, Pakistan during 2005 for promoting wheat growth under salt stressed conditions. Minimal salt medium containing ACC as sole nitrogen source was used for growth of rhizobacteria. Disinfected wheat seeds were germinated in petri plates. Uniformly germinated seeds were selected, inoculated with selected strains and placed between two filter paper sheets. Filter paper sheets containing seedlings were placed in jars. Jars were incubated at 25 ± 1 °C in the growth room under controlled conditions. Different salinity levels (1, 3, 5, and 7dS/m) for wheat were maintained by using NaCl. Hoagland solution was used to provide nutrients for growing wheat seedlings. The results revealed that inoculation with rhizobacteria containing ACC-deaminase showed significant increase upto 2.4, 1.9 and 5.0 folds in root elongation; shoot length and seedling fresh weight over uninoculated control, respectively. ACC11 and ACC16 were the most effective rhizobacterial strains which were further tested for assessing growth promotion phenomena of rhizobacteria under saline conditions. Results showed that there were 1.0 and 2.6 folds increase in root and 4.0 and 6.8 folds increase in shoot length with ACC16 and ACC11 over uninoculated control at higher salinity level (7.0 dS/m), respectively. At EC 3 dS/m, ACC11 increased fresh weight upto 1.2 times over uninoculated control under saline conditions. The results suggested that rhizobacteria containing ACC-deaminase activity could be used as successful inoculant for improving growth of wheat seedlings under saline conditions.
