- Abstract
This finding highlights the important mechanistic role of this regulatory site and the flexibility of ERAP1 as a drug target for manipulating the cellular immunopeptidome. Our cover shows a representation of the immunopeptidome of melanoma cells as it is altered by an allosteric inhibitor of ER aminopeptidase 1 (ERAP1), an intracellular enzyme that helps generate peptides for presentation by MHC-I molecules and regulates adaptive immune responses. Using an inhibitor targeting this allosteric regulatory site of ERAP1, Temponeras et al. discovered that it is possible to alter the repertoire of presented peptides in a unique manner, starkly different compared to the genetic downregulation of the enzyme.
