- Abstract
Different solvents were used to extract Inonotus hispidus and then the resultant extracts were given to cyclophosphamide induced immunosuppressed mice by gavage respectively. The extracts included petroleum ether extract at high (12 mg·kg-1) and low (6 mg·kg-1) doses, ethyl acetate extract at high (17 mg·kg-1) and low (8.5 mg·kg-1) doses, n-butanol extract at high (45 mg·kg-1) and low (22.5 mg · kg-1) doses, water extract at high (600 mg·kg-1) and low (300 mg · kg-1) doses, and purified ergosterol at high (10 mg·kg-1) and low (5 mg·kg-1) doses. Mice in different treatment groups were measured for immune organ indices, phagocytic index, phagocytic coefficient, ear swelling degree, ear swelling rate, and contents of cytokines and immunoglobulins. Hematoxylin-eosin staining was used to observe the morphology of spleen, thymus, pancreas and ileum tissue of mice under different treatments. The results showed that thymus index was significantly increased in the high dose petroleum ether extract group, the high dose water extract group and the purified ergosterol groups as compared with the model group. Both phagocytic index and phagocytic coefficient were significantly increased in the purified ergosterol groups, and the water extract increased phagocytic coefficient. The purified ergosterol and water extract also extremely significantly increased ear swelling degree and ear swelling rate. The content of IL-1β was significantly increased in the petroleum ether extract, water extract, ergosterol, and the high dose ethyl acetate extract groups. Similarly, the content of IL-2 was significantly increased in mice treated with the petroleum ether extract, ethyl acetate extract, water extract, and ergosterol. Except the low-dose n-butanol extract group, all the other extract groups and the purified ergosterol groups showed a significant increase in IFN-γ and IL-10 contents. The content of IL-4 was extremely significantly increased in mice treated with the petroleum ether extract, water extract, ergosterol and high dose ethyl acetate extract. The petroleum ether extract, water extract and ergosterol also significantly increased the content of IgG. The extracts of I. hispidus showed a good immune protection effect on cyclophosphamide induced immunosuppressed mice, and thus provided a reference for the further development of I. hispidus.
