- Abstract
Objective: To study the regulation of Zhenjingwendan decoction on corticosterone and neuron-specific enolase in serum and the expression of ORX receptor orexin1 receptor (OX1R) and orexin2 receptor (OX2R) in cortical limbic system of rats with post-traumatic stress disorder. Methods: PTSD model rats were established by single prolonged stress. The changes of OX1R and OX2R in brain region of rats were analyzed by real time PCR technique, and the changes of corticosterone and neuron specific enolase in serum were analyzed by Elisa. Results: The Zhenjingwendan decoction group increased the expression of OX1R and OX2R genes in the prefrontal cortex (PFC) and amygdala (AMY), reduced the levels of corticosterone and neuron-specific enolase in the serum, with significantly difference between the two groups (P<0.01). At the same time, the gene expression of OX1R and OX2R in hippocampus (HIP) was increased slowly. Conclusion: Zhenjingwendan decoction can regulate the content of corticosterone and neuron-specific enolase in rat serum and OX1R and OX2R receptors in ORX system, improve the imbalance of brain excitation-inhibition function, and alleviate the pathological state of insomnia and susceptible to fright effectively.
