GENOTOXIC AND CYTOTOXIC EFFECTS OF ASPIRIN IN WHITE MICE.

- Abstract
This study was conducted to estimate a genotoxicity and cytotoxicity of aspirin, using Swiss white mice Muss musculus. Three doses of aspirin were used 25, 40 and 50 mg. kg-1 bwt. based on cytogenetic endpoint (Chromosomal abnormalities in somatic bone marrow cells) and molecular cytogenetic technique, the Single Cell Gel Electrophoresis (SCGE) for Determination of DNA damage. The results of this study shown that aspirin had aneugenic and clastogenic effects in bone marrow cells of male white mice due to elevate mean differences ofmean value of numerical chromosomal abnormalities, which reached 4.40 ±0.58, 6.00 ± 0.58, 8.20 ± 0.58 and high value of mean difference of structural chromosomal abnormalities with thedose 50 mg.Kg-1 of aspirin, 28.40 ± 0.77 when gaps included and 24.60 ± 0.85 when gaps excluded. The Comet assay showed that the tested doses of aspirin induced DNA damage. The mean differences of mean values of bone marrow cells with damagedDNAwere 7.20 ± 0.65 with the dosage 25 mg.kg-1 of aspirin, 14.40 ± 0.65 with40 mg.kg-1, and20.40 ± 0.65 with 50 mg.kg-1. The mean differences ofmean values of kidney cells with damage DNA were 9.80 ± 0.61 25 mg.Kg-1, 15.00 ± 0.61 with40 mg.kg-1, and 22.00 ± 0.61 with 50 mg.kg-1 of aspirin and of liver cells were 9.69 ± 0.69 25 mg.Kg-1, 15.60 ± 0.69 with40 mg.Kg-1 and 21.80 ± 0.69 with 50 mg.Kg-1 of aspirin. Results of the current study suggest that the tested doses of aspirin, that induce different types of chromosomal abnormalities in bone marrow cells and DNA damage in bone marrow, kidney and liver cells may have genotoxic and/or cytotoxic effects in white mice.
