- Abstract
One of the theory of atherosclerosis is the theory of oxidative stress. The crucial role in the development atherosclerosis has endothelial dysfunction-term signifying decreased endothelial nitric oxide NO) produced from endothelial NO synthase. Decreased NO production may be caused by many risk factors:smoking, hypertension, diabetes. Excess formation reactive molecules such as reactive oxygen species (ROS) or reactive nitrogen species (RNS) in circumstances of decreased of NO bioavailability-oxidative stress, can stimulate oxidation of low density lipoprotein (LDL). Oxidized LDL is recognized by scavenger receptors in macrophages leading to foam cells formation, also influences reveral processes involved in atheroclerosis including leukocyte attachment, chemotaxis and expression of growth factors. Reactive species can be decreased or eliminated by antioxidant mechanisms. However, many studies of primary and secondary prevention of atherosclerosis failed to provide to support for use of antioxidants-vitamins C and E and beta-carotene.
