Optimization of Conditions for Phospholipase C by Bacillus sp. Z-13.

- Abstract
Taking the biomass and production of phospholipase C as detecting indice, the optimal conditions for phospholipase C of Bacillus sp. z-13 were conformed by single-factor experiments and uniform design experiments. The optimal conditions were as follows: Konjac powder 2.5 g/L, Soybean flour 30 g/L, K2 HPO4 ⋅ 3H2 O 2.8 g/L, Zn2 SO4 ⋅ 7H2O 1.3 g/L, inoculation size 0.5% rotating speed 150 r/min, culture temperature 32 °C ,original pH 9.0 and culture time was 15 h. In the optimal conditions, the living cells could reach up to 4.5 x 109 CFU/mL and sedimentary circle (milky white spot) diameter could reach up to 30 mm by egg yolk cylinder plate method.
