- Abstract
Aiming to shorten the detection period of Cronobacter and improve the detection accuracy, a rapid detection method for Cronobacter in infant formula milk powder was established in this study. The addition of promoter, inhibitor and repair agent for Cronobacter were screened by single-factor experiments, rapid growth medium )RGM) for Cronobacter was prepared, and the specificity and bacterial enhancement effect were further validated. The results showed that the rapid growth medium (RGM)was prepared with 5.0 g/L isomaltose as promoter, 3.0 g/L sodium pyruvate as repair agent and 5 mg/L vancomycin as inhibitor in buffered peptone water (BPW). While the initial inoculum of Cronobacter was 1 CFU, the concentration of bacterial solution could reach 109CFU/ml after culture at 36 ℃ for 24 h in RGM. For infant formula milk powder without and with probiotics, the proliferation rate of Cronobacter in rapid growth medium was 2 times and 10-20 times that of BPW, respectively. The specificity and enrichment effect of the detection method was good, and the bacterial concentration could be detected at 1 CFU, which could be used for the detection of Cronobacter in commercial infant formula milk powder.
