- Abstract
An analytical method was established for the determination of azithromycin in compound feed by ultra performance liquid chromatographytandem mass spectrometry (UPLC-MS/MS). The samples were extracted by H2O and acetonitrile, and NaCl was used as salting-out agent in this experiment. Extraction liquid was purified by dispersivesolid phase extraction with PSA and -NH2 sorben, and finally detected by UPLC-MS/MS in multiple reaction monitoring mode (MRM) via positive electrospray ionization (ESI), using 0.1% formic acid containing 5 mmol/L ammonium acetate as mobile phase,matrix standard curve quantification.The results showed that the correlation coefficient of the standard calibration curves for azithromycin in the concentration range of 0~50.0 μg/L was above 0.999. The limit of quantification was 2.0 μg/kg. The blank samples were spiked at three levels (2.0、4.0、8.0 μg/kg), and the average recoveries were between 75%~90% (n= 6), and the relative standard deviations were no more than 8.7%. It indicated that the method is simple, rapid and accurate; it is suitable for the determination of azithromycin in compound feed.
