- Abstract
Objective To study the effect of Bushen Zhuyun prescription on endometrial placenta protein 14 (PP14) and uterine natural killer (uNK) cells during the window of implantation (WOI) in superovulation mice, and to explore its immune mechanism in improving embryo implantation. Methods Sixty female Kunming mice with regular estrous cycles were randomly divided into a normal group, a model group, and a Bushen group. Embryo development and implantation were observed on the 8th day of pregnancy; the levels of the endometrial uNK cell marker CD49b+ and peripheral blood CD4+T and CD8+T cells were detected by flow cytometry, the serum estradiol (E2 ) and progesterone (P) levels were detected with radioimmunoassay, and the expression of endometrial PP14 was detected by immunohistochemistry. Results Compared with the normal group, the pregnancy rate, the average number of implanted embryos, the endometrial thickness, the number of glands, the expression of CD49b+ uNK cells and PP14, the levels of serum E2 and P, and the level of peripheral blood CD8+T cells were decreased in the model group (P < 0.05). Compared with the model group, the pregnancy rate and the average number of embryos implanted were increased in the Bushen group (P < 0.05). The endometrial tissue structure, thickness and number of glands, the expression of CD49b+uNK cells and PP14, serum E2 and P levels, and peripheral blood CD8+T cell levels were significantly increased (P < 0.05). In addition, there were no significant changes in the rate of vaginal plug formation or peripheral blood CD4+T cells in each group (P > 0.05). Conclusion Bushen Zhuyun prescription improves embryo development and implantation in superovulation mice by regulating WOI endometrial immune factors and receptivity.
