- Abstract
The article presents a novel proteomics method based on kinobeads for quantifying on- and off-target effects of kinase inhibitors in vivo. Kinobeads display multiple kinase inhibitors and preferentially bind kinases and other purine-binding proteins from cell lysates. The authors can determine affinity binding constants for hundreds of kinases from multiple cell lines and tissues through quantitative mass spectrometry to monitor binding of specific protein to the beads as concentrations of a free kinase inhibitor are increased.
