- Abstract
The aim of this study is to investigate the cytotoxic action and ameliorating effect on thefree radicals as reactive oxygen species (ROS) for well- known plants affron Crocus sativus. The ROS induced by cisplatin. the cytotoxicity ofcrude aqueous extract of saffron, cisplatin and a combination of them was measured onMDCK cell line at 24 and 48 hours incubation periods. The cells of MDCK were seeded in 96-well plate. Saffron and cisplatin wereused in a six serial dilutions (7.5, 15, 30, 60, 120, 240)μg/ml of each alone and fixed concentration (240 μg/ml) of cisplatin with the same serial dilutions of Saffron in combination together. The MTT(3-(4,5-Dimethylthiazole-2-yl)-2,5-diphenyl-2H-tetrazolium bromide) assay was used to determine the cells viability and the color intensity was measured by ELISA reader. The combination of saffron and cisplatin after 24 and 48 hours of incubation period showed that there was a significant decreasein the viability percent (P ≤ 0.05) for all concentrations in comparison with control group. The ROS measured by spectrophotometer after 48 hours incubation period, the results showed that there was no significant increase in the ROS values (P≤0.05)for higher concentration in combination (240+30, 240+60, 240+120, 240+240)μg/ml in comparison with control group.
